Alpha-synuclein is a 140 amino acids protein primarily expressed in the mammalian brain. It is mainly located at the synaptic end of neurons, where it plays a role in the binding of neurotransmitter-containing vesicles to the membrane and their release outside the cell. It also plays a role in controlling energy production in neural mitochondria. The normal form of the protein is a monomer. However, this protein can form pathological oligomers in diseases such as Parkinson’s disease or Lewis body atrophy coupled with neurotransmitters deregulation and inhibition of mitochondrial function. The presence of alpha-synuclein in blood raises the possibility of using the protein as a marker for disease states and to test the effect of drugs on neuronal activity. This kit has been designed for the detection and quantification of alpha-synuclein in serum, plasma, and cell culture supernatants.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
